The GLP-1 class mimics incretin hormones that boost glucose-dependent insulin secretion, suppress glucagon, slow gastric emptying, and increase satiety. The reference pharmaceuticals below are approved medicines; their manufacturer titration schedules are shown for educational comparison. Doses are escalated gradually to limit gastrointestinal side effects.
Semaglutide — reference: Wegovy (weekly)

At least 4 weeks at each step; 1.7 mg may serve as maintenance if 2.4 mg isn't tolerated. The type-2-diabetes product (Ozempic) tops out at 2 mg.
Tirzepatide — 'GLP-2 T', reference: Zepbound / Mounjaro (weekly)

A dual GIP + GLP-1 receptor agonist. Approved maintenance doses are 5, 10, or 15 mg — the lowest effective dose is preferred.
Retatrutide — 'GLP-3 R' (investigational)
A triple GIP + GLP-1 + glucagon receptor agonist. It is NOT approved by the FDA or EMA — all use is research-only. Phase 3 TRIUMPH trials studied 4 mg, 9 mg and 12 mg weekly, with roughly 28% average body-weight reduction at 12 mg over 68 weeks. There is no approved consumer titration schedule.
Missed-dose guidance (per approved labels)
- Semaglutide: windows vary by product — Ozempic can be taken within 5 days of the missed day, otherwise skipped; Wegovy's window is shorter. Then resume the normal weekly schedule.
- Tirzepatide: a missed weekly dose may be taken within 4 days (96 hours); after that, skip it and resume the usual day.
